Mirko Signorile was born in Bari, Italy, on February 3, 1974, and has spent his entire career inside the Puglia jazz scene that produced him. He earned his piano diploma with the highest marks and honors from Bari's "N. Piccinni" Conservatory, then in the summer of 1997 attended the Siena Jazz seminars, studying under Franco D'Andrea, Enrico Pieranunzi and Enrico Rava and receiving a scholarship as the course's best pianist. His jazz training continued under fellow Puglia pianist Gianni Lenoci, a teacher who later became a recording partner as well.

Signorile made his debut as a leader in 2003 with In Full Life (Soul Note), a record that drew critical notice and featured Puglia-scene contemporaries trombonist Gianluca Petrella and trumpeter Fabrizio Bosso, both Discogs-credited on the session. A year later he took part in Live In Clusone, a crowded Soul Note live document also crediting saxophonist Gaetano Partipilo and trumpeter Giovanni Falzone. His 2005 trio record The Magic Circle, billed to the Mirko Signorile Synerjazz Trio, built on that early momentum; Italian Wikipedia credits it with a second-place finish in Musica Jazz's "Top Jazz" critics' poll for best new Italian talent. In 2009, Clessidra (Emarcy/Universal) was voted album of the year by the critics' outlet ArgoJazz, a result that led directly to Signorile winning the Italian Jazz Awards' "Best Jazz Act" prize the following year.

Alongside his own-name records, Signorile has run several parallel projects. With saxophonists Gaetano Partipilo and Raffaele Casarano he co-leads Puglia Jazz Factory, documented on From The Heel (2012) and African Way (2015). With vocalist Giovanna Carone he has built a sustained body of work in Yiddish-language song, running from Betam Soul (2009) through Far Libe (2012), Mirazh (2014) and, in 2022, the orchestral commission Yitria with the Orchestra Giovanile della Valle d'Itria and guest Daniele Sepe. His more experimental side surfaced on 2016's Banaba, an electro-jazz project made with 99 Posse's Marco Messina that also carries a Discogs-credited guest trumpet spot from Paolo Fresu. Fresu later wrote the liner notes for one of Signorile's best-known group records, 2017's The Three Pianos, a three-way session with fellow pianists Claudio Filippini and Giovanni Guidi, released on Musica Jazz's own label.

That multi-pianist format returned in 2019's Playing Chess Keyboard (Dodicilune), a six-pianist collective session that reunited Signorile with his old teacher Gianni Lenoci alongside Domenico Cartago, Eugenio Macchia, Bruno Montrone and Danilo Tarso. The same year brought D'Amour, a duo record with Raffaele Casarano built around electronically treated saxophone and Fender Rhodes. Italian Wikipedia's own discography of Signorile also lists a 2010 trio album on Stradivarius and a 2022 trio project with singer Eugenio Finardi and Casarano called Euphonia Suite; neither could be independently confirmed at the Discogs credit level in this profile's research (a same-titled Euphonia Suite release does exist on Discogs but is credited solely to Finardi, with no personnel listed). Signorile now teaches piano at the "G. Paisiello" Conservatory in Taranto.

Signorile's playing sits at the meeting point of composed, cinematic melody and improvised jazz, a mix AllMusic has described as blending classical form with cinematic evocation and jazz phrasing. His training under Franco D'Andrea, Enrico Pieranunzi and Enrico Rava shows in a touch that favors clarity and structure over density, whether on the unaccompanied Soundtrack Cinema (2015) or the more open, electronics-inflected Banaba (2016). His catalog spans a wide range of settings by design: straight-ahead trio and quartet records for Auand's dedicated "piano series," multi-pianist summit projects (The Three Pianos, Playing Chess Keyboard), Yiddish-language vocal collaborations with Giovanna Carone, and the funk-and-Mediterranean-inflected Puglia Jazz Factory co-led with Gaetano Partipilo and Raffaele Casarano. He has also scored for cinema, theater and contemporary dance alongside his recording career.
